It would have to be a toss-up between an old Tom Hanks movie called "Bachelor Party", or the Austin Powers movie "The Spy Who Shagged Me".
I have them both and they still crack me up everytime.
Jim Carrey's 'Bruce Almighty' at top of list.
But some very funny ones that speak to people past the blush of youth:

Airplane! I love that slapstick, and the usually serious actors acting really silly.

There are many funny movies that have short parts of good humor, but for the most humor throughout I'd have to agree with PrisHaff about Airplane.
